Maple-syrup urine disease (MSUD) is an inborn error of metabolism characterized by the inherited deficiency of the branched-chain-2-oxoacid dehydrogenase complex which is required in the metabolism of the branched-chain amino acids. The detection of allo-isoleucine allows for positive identification of patients with MSUD reducing the false-positive neonatal screening results. Chromatographic separation of allo-isoleucine is readily achieved in 11 minutes analysis time with the accurate quantification using same sample.
